This may have been addressed in the past, if so I missed it.
I like to think that the professor would enjoy playing the game, even though there are a few inconsistencies with the cannon.
Does anyone know if any of his ancestors or other relatives have given the game a try?
And if so, have they made any comments?

I think you mean his descendants
and not that I've heard although I expect they wouldn't talk about it considering how the estate guards the copyright and intellectual property. It's almost like a celebrity or other well-known person playing, you wouldn't want anyone to know about it because it would likely ruin the entertainment and escape value for yourself once everyone figured out who you are in-game.

I don't know about Tolkien's descendants playing but I've often commented to my friends in game that I bet Tolkien would be proud of this game. The developers and script writers have done a fantastic job of making it as true to Tolkien's lore as possible. But not only that. That thousands of people gather to share an enjoyment of something based on one man's narrative; that a virtual representation of his Middle Earth exists for us to roam his imagined landscapes and immerse ourselves in his story is a massive compliment to his life's work.
